The quick answer is YES and YES.  Loading and unloading of a ship
happens before ships go into hyperspace.  You can also have a ship
unload, load something else(or the same thing, but why?) and still
move to another world.

The timing for using a route is a little different.  Loading happens
after most orders are processed, but still before going into
hyperspace(this stuff is in the sequence of play section of the
rules).  When a ship with a route and option AUTOUNLOAD set arrives at
its destination, it unloads on the same turn but after the production
phase as already occured.  Once you have a route set-up, you no longer
have to issue the load, send, and unload orders.  Just make sure the
ships are available for the automatic routing function to take over.
